Colossal Biosciences has announced the successful genetic engineering of the first dire wolves in over 10,000 years. The company, founded in 2021, used ancient DNA samples to decipher the dire wolf genome and rewrite the genetic code of the common gray wolf. The project resulted in the birth of three pups, Romulus, Remus, and Khaleesi, who were gestated by domestic dog surrogates.
The dire wolf, which went extinct 10,000 years ago, once roamed an expansive range from Canada to Venezuela. By making 20 specific edits across 14 genes in the gray wolf, scientists were able to replicate physical traits such as the dire wolf’s larger size, white coat, and powerful build. The resulting animals exhibit wild lupine behavior, maintaining distance from human handlers.
Colossal Biosciences aims to use these de-extinction techniques as a tool for conservation. Chief science officer Beth Shapiro suggests that the technology could help preserve endangered species by engineering more robust populations capable of surviving climate change. The company’s broader goals include the potential restoration of the woolly mammoth, the thylacine, and the dodo, following their recent creation of a chimeric woolly mouse.
Despite the scientific achievement, the project has faced criticism from the bioethics community. Experts like Robert Klitzman warn of the risks associated with cloning, including potential organ defects, premature aging, and the physical toll on surrogate mothers. There are also broader concerns regarding the potential for unintended consequences when introducing genetically altered species into modern environments.
During a recent observation, handlers noted that the dire wolf pups demonstrated unique vocalizations. When a technician sang, the pups responded with a howl, a sound that had not been heard on Earth for millennia. Matt James, the company’s chief animal officer, described the moment as a chilling confirmation of the project's success in reviving ancient traits.
The genetic modifications were based on DNA extracted from ancient remains, including a 13,000-year-old tooth and a 72,000-year-old ear bone. These samples provided the necessary blueprint for the painstaking lab work required to recreate the species. The company maintains that its efforts are a moral imperative to reverse the environmental damage caused by human activity.
As the pups continue to grow, Colossal Biosciences plans to monitor their health and development closely.
